RNA-based therapies offer a wide range of therapeutic interventions including the treatment of skin diseases; however, the strategies to efficiently deliver these biomolecules are still limited due to obstacles related to the cellular uptake and cytoplasmic delivery. Herein, we report the synthesis of a triggerable polymeric nanoparticle (NP) library composed of 160 formulations, presenting physico-chemical diversity and differential responsiveness to light. Six formulations were more efficient (up to 500 %) than commercially available lipofectamine in gene-knockdown activity. These formulations showed differential internalization by skin cells and the endosomal escape was rapid (minutes range). The NPs were effective in the release of siRNA and miRNA. Acute skin wounds treated with the top hit NP complexed with miRNA-150-5p healed faster than wounds treated with scrambled miRNA. Light-activatable NPs offer a new strategy to topically deliver non-coding RNAs. 相似文献

This work reports for the first time the nutritional profile, including proximate chemical composition, amino acids, fatty acids and minerals of Parastichopus regalis from the Mediterranean Sea (SE Spain). The studied species had a high moisture content, moderate protein and low lipid levels. The most abundant amino acids were glutamic acid, arginine and tyrosine. Polyunsaturated fatty acids, especially arachidonic acid, dominated the fatty acid profile. Iron, sodium, calcium and zinc were the most abundant mine rals. In general, P. regalis has a balanced nutritional quality suitable for human consumption. 相似文献

A new sol-gel application for the development of SPME fibers is described. Phenyltrimethoxysilane (PTMOS) and methyltrimethoxysilane (MTMOS) were the sol-gel precursors used at different proportions, together with different water contents, catalyst and reaction time. It was observed that obtaining a good film quality was determinant for a good extracting fiber performance. The film thickness ranged 0.2-1 microm and could not be increased by multi-coating processes. Apparently, a dense, non-porous microstructure was obtained. These coatings exhibited a strong hydrophobic character, as shown by the capability of extraction of long chain and apolar aromatic compounds, which, was comparable to that of the 100 microm polydimethylsiloxane (PDMS) and 65 microm carbowax-divinylbenzene (CW-DVB). The developed fiber has shown high thermal (350 degrees C) and organic solvent stability (ethanol, toluene and dichloromethane), thus bearing adequate characteristics to be associated to GC and potentialities that may also envisage suitability for HPLC. The new fibers may be useful for the microextraction of non-polar compounds, although at trace levels and in simple matrixes only, due to the susceptibility to competition. 相似文献

To a domain with conical points Ω, we associate a natural C*-algebra that is motivated by the study of boundary value problems on Ω, especially using the method of layer potentials. In two dimensions, we allow Ω to be a domain with ramified cracks. We construct an explicit groupoid associated to ?Ω and use the theory of pseudodifferential operators on groupoids and its representations to obtain our layer potentials C*-algebra. The solid-liquid phase diagram of the (1-ethyl-3-methylimidazolium bis{(trifluoromethyl) sulfonyl} amide + benzene) system was determined and allowed us to identify and characterize an equimolar inclusion compound, [emim][NTf2].C6H6, with a congruent melting temperature: this type of behaviour, reported here for the first time, together with the X-ray structure of the inclusion compound lays emphasis upon the interactions that are responsible for the existence of liquid clathrates at higher benzene concentrations. 相似文献
